Masks in substance painter are a very wide usage tool. The most common use is obviously using a mask to seperate different materials or effects from another part of the project, though you can use them for much more than this.
After you have laid down a base material you can instantly start using masks to apply effects like smart masks, generators and grunges. These create the desired effect to your model but you do have the option to take them further manually, using a brush you can work into these pre-generated masks and add some more uniqueness and realism to your model.
A benefit of using masks is the ability to use things like levels, invert mask and blur to adjust the mask to create some really nice effects onto your textures, I find that using these on masks looks a lot better than placing them on drawing layers.
